Ligands of the agarose derivatives described below are attached to CNBr-activated beaded agarose (Sepharose* 4B). Also, ligands are attached to the hydrocarbon spacer arms ethylenediamine succinic acid and 3,3'-diaminodipropylamine succinic acid which extend the ligands approximately 13 A and 21 A, respectively, from the agarose matrix. The spacer arm is attached to the agarose before coupling of the ligand. In the following descriptions:
DAPSA = 3,3'-diaminodipropylamine succinic acid.
LIGAND DENSITY.
The amount of coupled ligand is given in mg or µmole per ml of packed gel.
PACKAGING AND STORAGE.
Unless otherwise indicated, all agarose derivatives are supplied as packed gel in the storage buffer 0.02 M NaOAc pH 5 containing 0.01% NaN3 as preservative. The gel should be stored at 2° to 4°C in closed containers. Before use the gel should be washed with buffer of choice. After use the gel should be washed with storage buffer and refrigerated. For long term storage, the gel may be kept at -20°C after suspending in 50% (V/V) glycerol.
STABILITY.
The agarose derivatives will retain approximately 90% of ligand potency during a 12 month period when stored as indicated above. To insure optimum matrix characteristics and ligand potency; do not heat above 45° C, do not freeze without suspending in 50% (V/V) glycerol, do not bring to dryness and do not lyophilize without inclusion of a suitable stabilizer such as sucrose.
